Controlled Measurement Buffer for RT-DC Experiments

CellCarrier – Defined viscosity, osmolarity and pH for reproducible cell mechanics

CellCarrier is the standardized measurement buffer for real-time deformability cytometry (RT-DC).
It ensures controlled hydrodynamic stress conditions by providing defined viscosity, osmolarity and pH values — essential for reliable and reproducible mechanical phenotyping.

Sterile, batch-controlled measurement buffers with defined viscosity, osmolarity and pH for reproducible RT-DC experiments.

Why the Measurement Medium Matters

In RT-DC experiments using the AcCellerator platform, mechanical forces are defined by flow velocity and viscosity of the surrounding liquid.

CellCarrier is specifically formulated to create stable and reproducible hydrodynamic conditions inside the microfluidic channel.

Its composition is based on:

  • Sterile filtered PBS

  • < 1% methyl cellulose for viscosity adjustment

  • pH 7.4

  • Osmolarity 310–330 mOsm/kg

Each batch is quality-controlled and delivered in sealed, sterile flasks.

By standardizing the measurement environment, CellCarrier enables consistent comparison of datasets across experiments and laboratories.

CellCarrier Variants

Different experimental setups require different viscosity conditions.

CellCarrier

Designed for most mammalian cell lines and primary cells under standard RT-DC conditions.

CellCarrier B

Formulated for:

  • Smaller cells (e.g. blood cells)

  • Cells with higher stiffness

  • Channels with specific geometric constraints

CellCarrier B provides increased viscosity to ensure sufficient deformation contrast under defined flow conditions.

Biocompatibility and Experimental Stability

CellCarrier formulations have been tested with a wide range of:

  • Mammalian cell lines

  • Primary cells

  • Blood samples

The buffer maintains cell integrity during short-term measurement procedures while enabling precise mechanical characterization.

Because hydrodynamic stress directly depends on viscosity, using a defined and standardized buffer is critical for:

  • Quantitative comparison of elastic moduli

  • Reproducibility across instruments

  • Reliable inter-laboratory studies
